Design for C.NEST and S.NEST

NEST Lid

The NEST Lid is specifically designed for C.NEST/S.NEST cell culture kit. Each Cell Culture Kit has one NEST Lid and one standard microplate, which are applicable for C.NEST/S.NEST microbioreactor to perform suction and expulsion pressure through the fluidic channels.

The Principle

The NEST Lid includes an airflow port (1) for positive and negative air pressure to flow into and out of the lid chamber, this allows reciprocal mixing and provides a constant supply of oxygen to the cell culture to maintain a healthy environment. The evaporation rate corresponds to the distance between the airflow port and the fluidic tube (2). Theoretically, the left fluidic tube will have a higher evaporation rate than the one on the right.​

​The NEST has a chamber design (3) with deflector plates inside with different sized pores (4). The left pore is the smallest to reduce the most critical evaporation rate, with the middle and right pores becoming gradually larger to balance airflow resistance and maintain a consistent evaporation rate across the plate. The patented design of the lid greatly reduces evaporation and edge effects in multi-well plates.

Minimizie Evaporation and Edge Effects in Multiwell Plates

Medium evaporation is a common problem when using microplates for high-throughput cell culturing. The design of C.NEST/S.NEST microbioreactor and patented NEST Lid used with multiwell plate can minimize medium evaporation and edge effects by providing stable and controllable cell culture conditions for multiwell plate culture when compared to a regular cell incubator.

C.NEST/S.NEST microbioreactor and NEST Lid minimize evaporation and provided a more homogenous evaporation distribution in static and mixing culture conditions, which helps researchers avoid any negative influences due to edge effects such as inconsistent media concentration, variation in temperature or oxygen level between wells in cell-based assays or cell culture.

1. C.NEST Cell Culture Kit

The C.NEST Cell Culture kit has one NEST lid and a one standard 24-well or 96-well microplates, which are applicable for performing suction and expulsion pressure through the fluidic channels in the NEST lid. The cell culture kit is a one-time use consumable to bring the bioreactor environment to 1,000 μL – 1,600 μL (24-well) or 150-200 μL (96-well) early stage in C.NEST system.

2. S.NEST Cell Culture Kit

The S.NEST Cell Culture Kit has one NEST Lid and one 24-well sensor plate with DO and pH sensors attached to the bottom of each well, enabling dissolved oxygen and pH value monitoring. The conventional 24-well plate with NEST lids can be used for reciprocating mixing culture on S.NEST system. The cell culture kit is a one-time use consumable to bring the bioreactor environment to 1,000 μL – 1,600 μL (24-well) early stage in S.NEST system.

3. C.BIRD Cell Culture Kit

The C.BIRD Cell Culture Kit contains a multiwell plate and a functional lid (C.BIRD Lid). C.BIRD Cell Culture Kit is a one-time use consumable to bring the bioreactor environment to 150 – 200 μL (96-well format) and 1,000 – 1,600 μL(24-well format) in C.BIRD system. The standard functional lids are applicable for Greiner 24-well plate (cat no. 662102), Eppendorf 96-well plate (cat no. 30730011), and Corning 96-well plate (cat no. 3599).
